中国人胃癌组织DNA中癌基因Ha—ras等位基因的缺失

摘要 本文应用C—Ha—ras—1为探针与14个中国人Ha—ras胃癌患者癌组织和癌旁正常胃组织经限制性酶BamHI酶切后的DNA进行瑟慎印迹杂交。结果发现1例6.3kb/7.8kb杂合子患者在癌组织中丢失了6.3kbHa—ras等位基因。另外在一个胃癌杂合子患者家系的限制性片段长度多态性(restriction fragment length polymorphisms, RFLPs)分析中,发现患者的一个女儿也丢失了一个Ha—ras7.4kb的等位基因。这一现象可能是有丝分裂不分离或者有丝分裂重组造成的。 In this study, the human c-Ha-ras-1 6kilobase pair recombinant clone(EcoRI fragment) was used as a probe. The DNAs from 14 gastric carcinoma patients who are 13 heterozygous and 1 homozygous for Ha-ras locus contain two or one Ha-ras related BamHI restriction fragment. Southern blotting results showed similar patterns of allelic restriction fragments in gastric carcinoma and normal gastrc tissue DNAs in 13 cases. But one heterozygous patient(6.3kb/7.8kb) loses 6.3kb allelic restriction fragment in gastric careinoma. In addition, we examined RFLPs of the Ha-ras oncogene in one pedigree with heterozygou gastric carcinoma patient. The results show that one daughter of the patient losed one Ha-ras allele in WBC DNA, The Ha-ras allele loss may result from a mitotic nondisjunetion or a mitotic recombination event.
