抗CEAMCAb-血卟啉偶联物的制备及对肿瘤细胞的杀伤作用

摘要 目前在光动力学疗法中用于诊断和治疗恶性肿瘤的光敏剂血卟啉,尽管对肿瘤组织有一定的亲和力,但因对肿瘤细胞的选择性不够强,对肿瘤周围及其它正常组织也有损伤。因此,更优良的光敏剂的研制急待解决。将血卟啉(HP)与抗肿瘤单克隆抗体相连制备成“免疫光敏剂”。 The immunopholosensitizer was prepared by conjugating Hematoporphrin with anti-CEA McAb. It It showed cytotoxicity to human colon carcinoma cell line SW1116 which was CEA positive in the presence of light, but no cytotoxicity to CEA-negative Hep-2 cell line. The cytotoxicity of immunophotosensitizer was much higher than mixture of anti-CEA McAb and Hematoporphyrin. This property clearly makes it possible to use lower dose of drug, and minimize undesired side effects.
