摘要
根据杂交水稻种子稻壳来自母本及SSR分子标记的共显性特点,提出了一种基于杂交水稻种子鉴别其亲本的分子检测技术。首先提取杂交水稻种子稻壳的DNA,获得母本的DNA指纹图谱,再结合杂种带型,推导出父本带型。这样只要建立已有亲本的DNA指纹图谱库,通过比对,就可以鉴别杂交水稻的亲本是已登记亲本还是新创亲本。以金优207及其亲本作为实验材料,选用5对SSR引物验证了上述检测技术。
According to the distinguishing features that seed husk has the same genotype as the female parent and SSR markers are co-dominant, a molecular technology was proposed to identify the parents of hybrid rice based on its hybrid seed. Firstly, the fingerprinting of the female parent was obtained from the husk of hybrid seed. The fingerprinting of the male parent could be deduced by comparison of the band patterns between the female and its F1 hybrid. On the basis of the existing fingerprinting bank of hybrid rice parents. It could be easy to identify whether a parent is a new one or not. We tested successfully the identification technology by using Jinyou 207 and its parents as the materials and 5 sets of primers.
