摘要
目的:观察干扰素和卡介苗对肝癌大鼠单核巨噬细胞释放TNF的影响。方法:以二乙基亚硝胺(DEN)诱发大鼠肝癌,正常大鼠和肝癌大鼠分别腹腔注射干扰素(IFNα-2b)、卡介苗(BCG)或二者联用。从各组正常大鼠和肝癌大鼠体内分离出高纯度的血单核细胞、肺、脾和腹腔内的巨噬细胞,体外分别与人肝癌细胞联合培养,以ELISA酶联反应分别测定培养液中的TNF(Tumornecrosisfactor)含量。从未用免疫制剂的肝癌大鼠体内分离出高纯度的血单核细胞、肺、脾和腹腔内的巨噬细胞,体外培养并受IFNα-2b、BCG或二者联合作用后,人肝癌细胞联合培养,同上测定培养液中的TNF含量。结果:IFNα-2b或BCG均可促进上述单核巨噬细胞(正常大鼠和肝癌大鼠,在体和离体)释放TNF,并均以两者联用的效果最佳,使四种单核巨噬细胞释放TNF的量为对照组的11~37倍不等。结论:肝癌病人进行免疫治疗时,应提倡两种免疫制剂联合应用。
Objective :To investigate the influence of IFNa-2b and BCG on the release of TNF by monocytes and macrophages in the rat with hepatoma. Methods :Hepatoma was induced in rats by administration of diethylnitrosamine(DEN).The normal rats and hepatoma rats were treated respectively in vivo by IFNa-2b.BCG or combination of both and the controls were not treated with any immune stimulants(IS).The peritoneal macrophages,lung and spleen macrophages,monocytes were isolated from normal and hepatoma rats(at 8th and 16th week)and cocultured with human SMMC-7721 hepatoma cell,the concentration of TNF in media were measured by ELISA method,the peritoneal macrophages,lung and spleen macrophages,monocytes were isolated from control hepatoma rats(at 18th week)and treated with IS in vitro,and cocultured with human SMMC-7721 hepatoma cell,TNF level in media were assayed. Results : Both in normal and hepatoma rats,in vivo and vitro,IFNa-2b and BCG enhanced monocytes and macrophages to release TNF,particularly the combination of IFN a-2b and BCG increased the production of TNF by 1.1~3.7 times. Conclusion :The combined application of IS to patients suffering from hepatoma should be advocated vigorously.
