穿龙薯蓣愈伤组织诱导及其高产系的筛选 被引量:1

摘要 以穿龙薯蓣不同外植体为材料诱导愈伤组织并进行高产无性系的筛选,结果表明:种子是诱导愈伤组织的最佳外植体,最佳诱导培养基为MS+6-BA2.0mg/L+NAA2.0mg/L+2,4-D2.0mg/L,诱导率最高可达80.8%,愈伤诱导过程中没有出现褐化,愈伤组织生长迅速,平均每天可以增殖4.7mg。随着继代次数的增加,愈伤组织的生长速度增加,继代4-5次后生长最快,最高可达9.33mg/d。不同无性系及同一系在不同继代过程中都表现出差异,经过筛选,得到稳定的高产无性系8-31121和8-31123。 Different explants were induced and steady high production clone lines were selected in the paper. The results showed that seed was the best explant for inducing callus. The best culture medium was MS + 6 - BA 2.0 mg/L + NAA 2. 0 mg/L + 2,4 - D 2.0 mg/L, and the highest induction rate was 80. 8%. There was not browning in the process of inducing. Callus grew rapidly and the weight could increase 4. 7 mg/d on average. The growth speed of callus increased when the subculture times increased. Callus grew fastest after subculture 4 or 5 times and highest speed could reach 9. 33 mg/d. There were differences among different lines and the same line in different subculturing process, after screened, two steady high production clone lines were obtained, and they were named 8 - 31121 and 8 - 31123.
