中度嗜盐菌产α-淀粉酶发酵条件优化和酶活性质研究 被引量:4

摘要 实验室从新疆盐湖分离得到1株产α-淀粉酶中度嗜盐菌Bacillus sp.XJ1-05,对该菌株进行发酵条件优化和酶活性质研究,发现该菌株适宜的液态发酵工艺条件为:培养基碳源为酵母浸粉,氮源为大豆分离蛋白,两者比例1:1,培养温度33%,发酵时间72h,初始pH值7.0。最大酶活为98U,最佳反应温度为75℃,pH值为6.0-9.0。 The moderately halophilic bacteria Bacillus sp. XJ1-05 producing α-amylase was isolated from the salt lake of Xinjiang by the lab. The liquid fermentation conditions for α-amylase-producing and enzymology properties were studied. It was indicated that the optimal fermentation conditions were as follows: the carbon source was yeast extract; the nitrogen source was bean separation proteinum; the ratio of carbon source and nitrogen source was 1:1; initial pH7.0; 33℃ for 72h. The maxiumum enzymatic activity was 98U, the optimum temperature and pH of the enzyme were 75℃ and 6.0-9.0 respectively.
