maspin mRNA和蛋白在胃癌中的表达及意义

The clinical-pathogical significance and the expression of maspin mRNA and protein in human gastric carcinoma

目的:研究胃癌及正常胃黏膜中maspin的表达,并探讨其与临床病理特征的关系。方法:应用免疫组化S-P法检测胃管状腺癌(30例)、胃印戒细胞癌(30例)、正常胃黏膜组织(20例)中maspin的蛋白表达水平;同时应用半定量逆转录酶链反应(RT-PCR)检测正常胃黏膜组织和胃癌组织中maspin mRNA的转录水平。结果:在胃管状腺癌、胃印戒细胞癌、正常胃黏膜组织中,maspin蛋白阳性表达率分别为50%、46.7%、90%。正常胃黏膜组织maspin mRNA相对表达量明显高于胃癌组织(P<0.01)。maspin蛋白的表达与侵润深度、淋巴结转移相关,而与肿块的大小和TNM分期无关。Maspin mRNA的水平和蛋白的表达影响胃癌的润深度、淋巴结转移,而对肿块的大小和TNM分期无影响。结论:maspin基因的蛋白和mRNA在胃癌中表达下调可能在胃癌的发生发展中起着重要的作用,maspin的表达可能对胃癌的侵润转移具有抑制作用。

Objective： To investigate the expression of maspin mRNA and protein in gastric cancer and the relationship between clnical pathological parameters. In order to explore their roles in tumorigenesis and progression of gastric cancer.Methods ： Immunohistochemistry SP method was used to detemine expression of maspin in gastric adenocarcinoma, signet-ring cell carcinoma and normal gastric mucosa. The reverse transcriptase PCR was carried out to detect the level of maspin mRNA from the above mentioned 30 gastric adenocarcinoma and 20 normal samples. Results：The expression rate of maspin protein was 50% in gastric adenocarcinoma , 46.7% in signet-ring cell carcinoma, and 90% in normal gastric mucosa , respectively. The relative transcription level of maspin mRNA in normal gastric mucosa was higher than that in gastric adenocarcinoma .Maspin protein expression was significantly related to the depth of invasion and lymph node metastasis of gastric carcinoma, but did not relate to tumor size and TNM staging. The relative mRNA transcription level of maspin signifi- cantly correlated with the lymph node metastasis and the depth of invasion of carcinoma,but did not relate to TNM staging or tumor size. Conclusion： Down-regulated expression of maspin play an important role in gastric tumor. It may have inhibitory effects on invasion and metastasis of gastric tumor. Maspin might be objective indicator for pathobiological behaviors of gastric tumor.
