摘要
针对1,3-丙二醇发酵通常需要在完全密闭并充满氮气的环境下进行,且工艺操作复杂、生产成本偏高等问题,采用经过耐氧驯化的克莱伯氏菌ZU-05菌株,在非厌氧条件下利用甘油发酵生产1,3-丙二醇,并对主要发酵工艺参数进行了优化.摇瓶试验结果表明,在体积接种量10%,甘油20 g/L,玉米浆6 g/L,葡萄糖8 g/L,pH值6.7,37℃的非厌氧发酵条件下,甘油利用率为95%,1,3-丙二醇转化率可达51%.该研究结果对于简化1,3-丙二醇的发酵工艺、提高经济效益、促进该项技术的产业化进程等具有重要意义.
Considering that the current bioproduction of 1,3-propanediol is mainly carried out under strict anaerobic conditions, which requires airtight production equipment and large amount of nitrogen to maintain the oxygen free conditions in the fermentation system, and the production procedure demands quite complex operation technique which leads to high cost, an endure-oxygen Klebsiella pneurnoniae strain ZU-05 was used to produce 1,3-propanediol from glycerol under non-strict anaerobic conditions, and the main fermentation parameters were optimized. Under the non-strict anaerobic condition with 20 g/L glycerol ,6 g/L corn syrup, 8 g/L glucose and inoculation volume of 10% at pH 6.7, 37 ℃, the utilizing rate of glycerol and yield of 1,3-Propanediol were 95 % and 51% within 60 h, respectively. The results are meaningful for simplifying the procedure and increasing the profit of production from glycerol to 1,3-propanediol.
