促红细胞生成素对新生大鼠缺氧缺血性脑损伤凋亡的研究 被引量:2

摘要 目的研究单次注射外源性人重组促红细胞生成素(rhEPO)对新生大鼠缺氧缺血性脑损伤(HIBD)凋亡的影响,探讨rhEPO神经保护作用分子机制。方法新生7d龄Wistar大鼠,随机分为假手术组、HIBD组、rhEPO治疗组。采用Rice等方法建立新生大鼠缺氧缺血性脑损伤模型(HIBD模型),rhEPO治疗组于缺氧缺血完成后即刻腹腔内注射rhEPO5000U/Kg1次。各组分别于术后及给药后不同时间点断头取脑组织,HE染色观察神经细胞形态变化、用免疫组化方法检测凋亡相关蛋白:Bcl-2、Bax蛋白的表达。结果HIBD组神经细胞凋亡增加,与假手术组比较差异有统计学意义(P<0.01);rhEPO治疗组神经细胞凋亡减少,与HIBD组比较差异有统计学意义(P<0.01);Bcl-2蛋白在假手术组呈低水平表达,HIBD组表达增高,rhEPO治疗组则较假手术组及HIBD组表达更高;Bax蛋白在假手术组中表达弱,HIBD组表达增高,rhEPO治疗组则HIBD组表达低。结论rhEPO通过减少新生大鼠脑神经细胞坏死和凋亡来发挥其神经保护作用。
