HpD-PDT对人乳腺癌细胞增殖、凋亡的影响及作用机制

摘要 目的:研究血卟啉衍生物(hematoporphrphyrin derivative,HpD)介导的光动力疗法(photodynamic therapy,PDT)对人乳腺癌细胞MDA-MB-231增殖、凋亡的影响,探讨其可能的作用机制,为HpD-PDT治疗乳腺癌提供理论依据。方法:体外培养的人乳腺癌细胞MDA-MB-231分为4组:A组(空白对照)、B组(单纯激光)、C组(单纯光敏剂)、D组(激光+光敏剂)。应用流式细胞术分析HpD-PDT对细胞周期的影响,免疫组织化学技术检测HpD-PDT对PCNA、Bax和Bcl-2蛋白表达的影响。结果:HpD-PDT作用后,乳腺癌细胞内G0/G1期细胞比显著高于对照组(P<0.05);PCNA阳性表达率明显低于对照组(P<0.01);Bcl-2蛋白的阳性表达率明显下降(P<0.05);Bax蛋白阳性表达率各组间均无统计学差异;实验组HpD-PDT作用后Bax/Bcl-2比值增高,呈时间依赖性变化。结论:HpD-PDT的作用机制可能与抑制肿瘤细胞增殖,诱导细胞凋亡有关。 Objective: To evaluate the effect of hematoporphrphyrin derivative (HpD) mediated photodynamic therapy (PDT) on proliferation and apoptosis of human breast cancer cell line MDA -MB -231, and to investigate the mechanism of its action. Methods : MDA - MB - 231 cells were treated by 4 ways : group A ( control ), group B (laser), group C (HpD), group D (HpD -PDT). Human breast cancer cells (MDA -MB -231 ) were treated with HpD - PDT in vitro. Cell cycle analysis was measured by flow cytometry (FCM) and the expressions of PCNA, Bax and Bcl - 2 were detected by immunohitochemistry. Results : After HpD - PDT treatment, the cell number in G0/G1 phase increased (P 〈0.01 ) and in S phase decreased (P 〈0.05). The expressions of PCNA and Bcl -2 in the experimental group were obviously lower than that in the control groups (P 〈 0.05 -0.01 ). Bax protein expression was no differences among all groups ( P 〉 0.05 ). The ratio of Bax/Bcl - 2 in the experimental group was higher than that in the control groups ( P 〈 0.01 ) , and it was significantly raised along with the time prolongation after PDT action. Condution:The mechanism of HpD - PDT may be related to inhibit human breast cancer cell proliferation and induce cell apoptosis.
