摘要
目的通过转染野生型p53基因,并与5-氟尿嘧啶(5-Fu)联合作用,观察其对鼻咽癌细胞生长的影响。方法将克隆有野生型p53基因的pUHD10-3-p53质粒,用脂质体(lipofectamine)介导转染人鼻咽癌CNE2细胞,在培养液中加入5-Fu,用四甲基偶氮唑(MTT)法分析细胞生长情况。结果人鼻咽癌CNE2细胞加入含5-Fu的培养液后,第4天生长抑制率为54.85%;CNE2细胞转染wt-p53基因后,第4天生长抑制率为45.90%。人鼻咽癌CNE2细胞转染wt-p53基因后应用5-Fu处理,细胞的生长抑制率第4天可达到85.82%,其生长明显受到抑制。结论野生型p53基因能抑制CNE2细胞生长,wt-p53与5-Fu联合作用,对人鼻咽癌CNE2细胞生长有明显的抑制效果。
Objective To investigate the effect of combination of wild type p53 (wt-p530) gene transfected with and 5-fluorouracil (5-Fu) on the growth of nasopharyngeal carcinoma. Methods The nasopharyngeal carcinoma line CNE2 that transfected with plasmid pUHI)10-3-p53 recombined,with wt-p53 gene by lipofeetamine-mediate were cultured in medium contained 5-Fu. 3-(4,5-Dimethy-2-thiazolyl)-2,5-diphenyl-2H-tet- razolium-bromid (MTT) technique was used to analyse the growth of nasopharyngeal carcinoma line CNE2. Results The growth of nasopharyngeal carcinoma line CNE2 were inhibited after being exposed to 5-Fu,and 54. 85% of the cells were inhibited on the fourth day. After' tmnsfeeting with wt-p53 gene ,45. 90% of the nasopharyngeal carcinonma line CNE2 were inhibited on the fourth day. Affter the nasopharyngeal carcinoma line CNE2 that transfeeted with wt-p53 gene exposed to 5-Fu for 4 days, 85.82% of the cells were inhibited. Conclusion The growth of nasopharyngeal carcinoma was obviously inhibited by the combination of wild type p53 (wt-p53) gene and 5-fluorouracil (5-Fu).
