摘要
目的观察偏硅酸钠(Na2SiO3)对氧化低密度脂蛋白(ox-LDL)诱导的ECV-304细胞与人外周血单个核细胞黏附的影响,探讨其抗动脉粥样硬化(AS)作用的机制。方法ECV-304细胞分为5组:对照组、ox-LDL组、Na2SiO3高剂量组(硅68.5mg/L)、中剂量组(硅34.2mg/L)、低剂量组(硅17.1mg/L)。分离正常健康人外周血单个核细胞,计数与各组ECV-304细胞黏附数,检测细胞培养液中IL-8水平;采用流式细胞术测VCAM-1的蛋白表达量;半定量RT-PCR检测VCAM-1、IL-8mRNA的表达。结果Na2SiO3减少ECV-304单个核细胞黏附数(P<0.01),降低IL-8浓度(P<0.01),降低VCAM-1的蛋白表达量(P<0.05),下调VCAM-1、IL-8mRNA的表达(P<0.01)。结论Na2SiO3能够下调ox-LDL诱导的ECV-304IL-8和VCAM-1的表达。
Objective To observe the effect of Na2SiO3 on mononuclear cells adhesion to ECV-304 cells induced by oxidized low density lipoprotein(ox-LDL).MethodsLogarithmic growth phase of ECV-304 cells were divided into control,ox-LDL,high,middle and low dose(Si 68.5,34.2,17.1 mg/L)of Na2SiO3 groups.The level of interleukin-8(IL-8)in cell culture fluid was tested,the protein expression of vascular cellular adhesion molecule(VCAM)-1 was measured by flow cytometry,the gene expressions of VCAM-1 and IL-8 were measured by half quantitative reverse transcription polymerase chain reaction(RT-PCR).ResultsNa2SiO3 decreased the level of IL-8 and the number of mononuclear cell adhesion(P〈0.01),and the expressions of VCAM-1 protein(P〈0.05),VCAM-1 gene(P〈0.01)and IL-8 gene(P〈0.01).ConclusionsNa2SiO3 can inhibit the expression of adhesion molecule in ECV-304 cells induced by ox-LDL.
