通脉益智胶囊对VAD大鼠海马CA1区形态结构损害的干预作用

摘要 目的:通过观察通脉益智胶囊对血管性痴呆(VAD)大鼠学习记忆能力和海马形态学变化的影响,探讨其防治VAD的作用。方法:应用高脂血症大鼠,采用脑缺血再灌注的方法建立VAD动物模型,利用Y型电迷宫检验大鼠的学习记忆能力,使用透射电镜和光镜观察分析海马CA1区形态结构的变化。结果:电迷宫实验表明模型大鼠学习记忆能力明显下降,而给予通脉益智胶囊组学习记忆能力改善,与模型组相比有显著性差异。形态学观察显示模型组大鼠海马CA1区神经细胞排列紊乱、数目减少,部分神经细胞核深染、固缩。电镜下神经细胞核膜皱褶内陷,核固缩,染色质凝集、边集,细胞器排列紊乱。通脉益智胶囊组的神经细胞形态与模型组相比在光镜、电镜下均有明显改善。结论:通脉益智胶囊对VAD大鼠海马CA1区的神经细胞具有保护作用,从而提高了学习记忆能力。 Objective To explore the influence of Tongmaiyizhi capsule on ability of learning and memory and morphology of hippocampus in vascular dementia model rats. Methods Animal models with vascular dementia were prepared by cerebral ischemia-reperfusion at hyperlipidemia model rats. We observed the rats' ability of learning and memory in the y-type electric maze and the change of morphology of hippocampal CA1 sector by the TEM and light microscopy. Results The electric maze experiment showed that ability of learning and memory obviously decreased in model rats, nevertheless the change of ability of learning and memory in group with Tongmaiyizhi had significant effect compared with model rats. Morphological observation indicated that the nerve cell decreased in number, arranged in disorder, and partly nuclei deeply stained, degenerated, in CA1 sector of hippocampus of the model rats. TEM showed that for nerve cell,the membrane folds invaginated,the nuclei degenerated,the chromatin condensed and margined,and the organelles arranged in disorder. Under the TEM and light microscopy, nerve cell morphology of Tongmaiyizhi group had clear amendment compared with the model group. Conclusion Tongmaiyizhi capsule can protect the nerve cell in CA1 sector of hippocampus of the VAD rats, so it improves the ability of learning and memory.
