支气管哮喘患者外周血嗜酸粒细胞上前列腺素D2受体改变研究

摘要 目的了解不同病期支气管哮喘(简称哮喘)患者外周血中嗜酸粒细胞(EOS)上前列腺素D2(PGD2)受体(DP1/CRTH2)结合容量改变情况。方法随机病例对照研究。分为正常对照组24例,平均年龄(5.8±2.7)岁,哮喘急性发作期组18例,平均年龄(4.7±3.8)岁和缓解期组12例,平均年龄(3.9±3.1)岁。用放射配体结合分析法测定外周血EOS上DP1/CRTH2结合容量。结果哮喘急性发作期组和缓解期组患者EOS数显著增加(×10^6/L)(急性发作期组:297±58;缓解期组:157±34和对照组95±26)(P〈0.01),EOS上CRTH2受体结合容量和总结合容量比正常对照组和缓解期组显著增加(fmol/10^6细胞)(CRTH2分别为:245.58±48.27、154.13±67.25、119.29±53.47;DP总结合分别为:456.14±25.57、337.51±38.52、301.22±39.54)(P〈0.01)。结论哮喘急性发作期组CRTH2受体结合容量增加,PGD2激发的EOS的趋化反应将偏向于CRTH2受体中介的生物学效应。 Objective To investigate the change of prostaglandin De receptor (DP1/CRTH2) on eosinophil(EOS) in exacerbation and remission phase patients with bronchial asthma(asthma), Methods Randomized case control clinical trial was designed. Thirty patients were randomly divided into two groups: exacerbation phase group 18 cases(4.7±3.8) years, and remission phase group (3.9 ± 3.1 ) years, normal control was 24 cases(5.8±2.7) years. Radiological binding assay was used to study the DP1/CRTH2 receptors on EOS in peripheral blood. Results The EOS counts in exacerbation phase group were significantly higher than those in remission phase group and normal controls( 〉( 10^6/L) (exacerbation phase group,297±58, remission phase group 157 ± 34 normal controls 95± 26) ( P 〈0.01 ). The combining capacity of CRTH2 and the total combining capacity on EOS in exacerbation phase group were significantly higher than those in remission phase group and normal controls(fmol/10^6 cell)(245.58±48.27 vs 154.13 ± 67.25,119.29±53.47 and 456.14±25.57,337.51±38.52,301.22±39.54)( P〈0.01).Conclusions The combining capacity of CRTH 2 on EOS in exacerbation phase group are significantly higher, this imply that PGD2 mediates the chemotaxis of EOS will be via CRTH2 receptor.
