摘要 目的:研究脑损伤后神经元和神经胶质细胞内ApoE和S-100的表达随脑损伤时间变化的规律及其在脑损伤时间推断中的作用。方法:通过建立大鼠脑挫伤模型,利用免疫组化染色法(S-P法)对对照组及伤后不同时间组内脑细胞中的S-100和ApoE两种蛋白进行检测,并作统计学分析。结果:伤后0.5h组大脑皮质神经元中ApoE即出现显著表达(P<0.01),而S-100阳性细胞在伤后2h开始表达显著增强(P<0.01)。随着损伤时间的延长,ApoE和S-100阳性细胞数目及阳性表达范围逐渐扩大。伤后3d组ApoE阳性细胞表达数量及染色强度达到高峰,且分布广泛,随后表达下降;到5d组时,S-100阳性产物达到高峰,后下降,但仍高于对照组。结论:脑挫伤后S-100和ApoE在损伤局部的染色变化有一定的时间规律性,可以用于脑挫伤的时间推断。 Objective: To observe the correlated expressions of S-100 and ApoE after doing experiment which related to brain contusion with rats, as well as to research the changes between neurons and glial cells when brain was injured, after that to evaluate what role played in brain contusion. Methods: Through establishing model of brain contusion in rats, immunohistochemical method (S-P) was applied to detect the expressions of S-100 protein and ApoE in control group with different time after injury. The immunostaining results were measured with statistics analysis. Results: At 0.5h after injury, ApoE protein of neurons in the cerebral cortex had a positive expression, and the S-100 positive cells after 2 h of injury, expression increased powerfully. With the extension of injury, the number of positive cells and the scope of expression gradually expanded. ApoE expression of the number of positive cells and staining intensity reached a peak in Group 3 d after injury, and distributed widely, then the expression decreased. In Group 5d after injury S-100 positive product peaked and then have a significant drop, but still higher than that in control group. Conclusion: The regular changes of post injury S-100 protein and ApoE protein expressions in rats can be used in estimation of post brain contusion interval.
