臂丛神经根性撕脱伤后TGF-β、TNF-α、bFGF在脊髓中表达的变化

摘要 目的:通过观测大鼠臂丛神经根性撕脱伤后脊髓中运动及感觉神经元内转化生长因子β(TGF-β)、肿瘤坏死因子α(TNF-α)、碱性成纤维细胞生长因子(bFGF)的表达变化,探讨臂丛神经撕脱伤后神经相关细胞因子TGF-β、TNF-α、bFGF可能对神经再生的影响。方法:清洁级大白鼠54只,随机分为正常对照组和损伤术后1、3、7、10d和14d组,共6组。损伤组均从前路造成臂丛C5~T1神经根性撕脱伤。按各组所示时间取脊髓分别检测TGF-β、TNF-α及bFGF的蛋白表达。并通过观察阳性细胞数对组织切片中TGF-β、TNF-α及bFGF进行半定量分析及比较。结果:臂丛神经根性撕脱伤后,脊髓TGF-β、TNF-α、bFGF表达均增高,前两者3d时达到高峰,随后下降,14d时仍高于对照组。而bFGF7d时达到高峰,随后下降,14d时下降到接近正常水平。结论:臂丛神经根性撕脱伤后神经因子在中枢神经元表达增高,可能有保护神经元、促进神经再生的作用。 Objective: To investigate the variation of transforming growth factor (TGF-β), tumor necrosis factor (TNF-α), basic fibroblast growth factor (bFGF) expressions in spinal cord after root avulsion of brachial plexus. Discussion the effects of nerve-related cytokines-TGF-β, TNF-α, bFGF on nerve protection and regeneration after brachial plexus avulsion. Methods. Fifty-four rats were involved in this study and according to the observing time in 1 st, 3 rd, 7 th, 10 th, and 14 th day after avulsion, and the control were divided into 6 groups. The damage of avulsion groups caused by the previous group brachial plexus C5- T1 nerve root avulsion. The spinal cord from the group was got by the time. By immune histochemistry, the expression of nerve-related cytokines on spinal cord TGF-β, TNF-α, bFGF on the neurons of coresponding spinal cord was detected. Comparative and semi-quantitative analysis through observation on positive cells of TGF-β,TNF-α,bFGF in the biopsy. Results: After the root avulsion of brachial plexus occurred, expressions of nerve-related cytokines-TGF-β, TNF-α, bFGF all were increased, the first two reached to the peak at the 3rd day, then decreased, it was still higher than normal control until the 14th day. bFGF protein and one week at a peak and then decline, 14th d dropped to near normal levels. Conclusion. After root avulsion of brachial plexus, neurotrophic factors-TGF-β, TNF-α, bFGF expressions increase on target the neurons of corresponding spinal cord. It maybe the auto regulation and may protect neuron and improve nerve regeneration.
