摘要
目的采用HPLC—ELSD测定桑树不同各部位中的1-脱氧野尻霉素(DNJ)含量,并测定四川省4个代表性蚕桑基地的桑树各部位DNJ的含量。方法色谱柱为DIKMA Ineresil NH2 column(250mm×4.6mm,5μm);流动相为乙腈-甲醇-水(70:10:20);流速1ml·min^-1,柱温35℃。结果线性范围1.02~12.24μg(r=0.9995);平均回收率为98.7%(RSD=1.58%)。结论不同种不同产地桑树中DNJ含量差异较大,绵阳三台县和泸州泸县的山桑干皮含量最高(分别为8.145、7.624mg·g^-1),桑叶含量居中,桑椹含量为最低(0.345mg·g^-1)。
OBJECTIVE To establish HPLC - ELSD method and determine 1 - deoxynojinimycin(DNJ) in different parts of different varieties of mulberry. METHODS HPLC analysis was performed on a DIKMA Interesil NHz Columm (250mm × 4.6mm,5μm). The mobile phaseconsisted of aeetonitrile -methanol -water 170 : 10 : 20). The flow rate was 1.0ml·min^-1. The temperature was set at 30℃. RESULTS The HPLC - ELSD method could quantitatively determine DNJ in different parts of different varieties of mulberry. The linear range was 1.02 - 12.24 μg ( r = 0. 9995 ) and the average recovery was 98.7% with RSD of 1.58% . CONCLUSION The identification methods are reproducible and specific, the quantitative determination method is sensitive and accurate. There is great different of DNJ in difference parts in different varieties of mulberry. The contents of DNJ in M. bombycis Koidz form Lu town (Luzhou city) and Shantai town (Mianyang city)were 8. 145,7. 624 mg·g^-1, respectively. It is middle DNJ in mulberry leaf and minimum in mulberry fruit(the lowest: 0. 345 mg·g^-1).
