摘要
[目的]体外分离、培养奶牛乳腺成体干细胞(mammary stem cells,MaSCs),研究其生物学特性并对其进行扩增和鉴定。[方法]用贴壁筛选法纯化牛MaSCs,传代扩增,测定生长曲线并进行细胞核型分析。[结果]通过体外分离培养获得纯度较高的MaSCs,观察到MaSCs的生物学特性,细胞具有正常核型,分离得到的细胞确实为比较原始的干细胞。[结论]成功地建立了一种分离培养牛MaSCs的方法,分析MaSCs的生物学特性,为进一步深入研究MaSCs的诱导分化及其在乳腺生物反应器中的应用打下基础。
[Objective] The research aimed to isolate and culture cow mammary stem cells(MaSCs) in vitro and study the biology characteristics and make the amplification and identification.[Method] Cow MaSCs ware isolated and purified by adherence plasticity method.The growth curve was measured and the karyotype was analyzed.[Result] Higher-purity MaSCs were obtained by culture in vitro and the biology characteristics were observed. Cells had normal karyotype and the isolated cells were confmned as ~tively primordial stem cells. [Conclusion] A kind of method for the isolation and culture of MaSCs was successfully established and the biology characteristics of MaSCs were analyzed. These results laid the foundation for studying the induction 'and differentiation of MaSCs and their applications in manmary gland bioreaetor furor.
