鹅源H5N1禽流感病毒在感染雏鸡肾脏的分布及所致病理变化 被引量:3

摘要 目的研究鹅源H5N1亚型禽流感毒株人工感染雏鸡后,禽流感病毒在感染雏鸡肾脏内的分布及其所致的病理损伤变化。方法120只1d龄SPF雏鸡随机分为:对照组(C组)、7d龄和21d龄禽流感病毒感染组(Ⅰ、Ⅱ组)。I组和Ⅱ组雏鸡分别于7和21d龄时,分别经鼻、眼、口感染鹅源H5N1亚型禽流感病毒稀释液,3组雏鸡感染后1、3、4、5、7、14、21d,随机抽取5只,心脏采血处死,取肾脏,应用免疫酶组织化学和图像分析技术等手段检测禽流感病毒,并观察肾脏的病理变化。结果雏鸡感染鹅源H5N1亚型禽流感病毒后1~14d,其肾脏血管内皮细胞、肾小管上皮细胞内均可检测到病毒抗原,并呈现不同程度的病理形态损伤。结论鹅源H5N1亚型禽流感病毒对雏鸡肾脏具有较强的嗜组织特性,可造成雏鸡肾脏严重损伤。 The viral distribution and pathologic changes of kidneys of chickens experimentally infected with H5N1 subtype avian influenza virus were investigated in this study. One hundred and twenty one-day-old specific-pathogen-free chickens were divided randomly into three groups, group Ⅰ (the 7-day-old group), group Ⅱ (21-day old group) and control group(C group). Chickens in groupⅠ and Ⅱ were inoculated with influenza viruses on the 7-day-old and 21-day-old by nasal, eyes and oral route simultaneously. Five chickens from each groups were killed by heart bleeding at 1, 3, 4, 5, 7, 14 and 21days post infection. The virus distribution and pathologic changes of the infected chickens were found to be limited to kidney by the immunohistochemistry and image analytical technique. The viral antigens were detected on endothelial ceils of blood vessels and renal tubular epithelial cells, respectively. Furthermore, patho-morphologic lesion in the kidney was observed throughout the entire experimental periods. This study demonstrateds that HSN1 avian influenza virus had strong tropism for chicken kidney and could cause significant kidney lesions in the infected chickens.
