整合素αvβ_3、VEGF及MMP-9在膀胱移行细胞癌中的表达及意义 被引量:1

摘要 目的:检测整合素αvβ_3、血管内皮细胞生长因子(VEGF)及基质金属蛋白酶9(MMP-9)在人膀胱移行细胞癌的表达,探讨整合素αvβ_3与膀胱移行细胞癌血管生成及浸润转移的关系。方法:采用免疫组织化学SABC法检测59例膀胱移行细胞癌组织及15例正常膀胱黏膜组织中整合素αvβ_3、VEGF、MMP-9的表达情况。结果:59例膀胱移行细胞癌中αvβ_3、VEGF、MMP-9的阳性表达率分别为45.76%、64.41%和55.93%,15例正常膀胱黏膜组织中三者的阳性表达率分别为13.33%、6.67%和6.67%三种蛋白在膀胱移行细胞癌与正常膀胱黏膜组织间的表达阳性率差异均有统计学意义(P<0.05,P<0.01);αvβ_3、VEGF、MMP-9的表达与膀胱移行细胞癌的分级分期相关;αvβ_3、VEGF的表达与膀胱移行细胞癌的癌灶直径、单发/多发相关;αvβ_3的表达与膀胱移行细胞癌患者的年龄相关;整合素αvβ_3与VEGF及MMP-9之间的阳性表达均呈正相关(r=0.3703,r=0.3182,P<0.01)。结论:整合素αvβ_3在膀胱移行细胞癌中表达升高,并与分级分期等指标相关,表明其具有促进膀胱移行细胞癌生长的作用。整合素αvβ_3可能通过协同VEGF及MMP-9作用,促进膀胱移行细胞癌的血管生成及侵袭转移。 Objective:To study the expression of integrin αVβ3, vascular endothelial growth factor (VEGF), and matrix metalloproteinase-9 (MMP-9) in bladder transitional cell cancer, and to discuss how integrin αVβ3 play role in the angiogenesis, invasion and metastatic of BTCC. Methods:Using the immunohistochemical SABC kit to detect the expression of integrin αVβ3, VEGF, MMP-9 in 59 BTCC tissues and 15 normal bladder tissues. Results: The expression positive percentage of integrin αVβ3, VEGF, MMP9 in BTCC was 45.76%, 64.41%, 55.93%, and the expression positive percentage in normal bladder tissues was 13.33%, 6.67%, 6.67%. The difference between two group was significant (P〈0.05, P〈0.01). The expression of integrin αVβ3, VEGF, MMP-9 has re lation with stage and grade. The expression of integrin αVβ3 and VEGF has relation with simple/poly disease and diameter of tumor. The expression of integrin αVβ3 also has relation with age of patients. There is a positive corre-lation between the expression of integrin αVβ3 and VEGF or between integrin αVβ3 and MMP-9 (r=0.3703, r=0.3182, P〈0.01). Conclusions:The expression of integrin αVβ3 increased in BTCC, and has relation with the clini-copathlogieal index such as stage or grade, integrin αVβ3 might has cooperation with VEGF and MMP-9 in the angiogenesis, invasion and metastatic of BTCC.
