摘要
以鸡胚分离结合血清学鉴定,从近几年江苏省及河南省分离到来自鸡和鸽子的6株新城疫病毒(NDV)。病毒蚀斑纯化后,选取3个克隆利用RT-PCR技术扩增F基因重要功能区片段,在对PCR产物直接序列测定分析基础上,选取一致克隆株进行全长F基因克隆、序列测定分析。根据分离株序列与GenBank中NDV相关毒株序列比较分析,结果表明,所分离的6个分离株归属为3个NDV基因型,其中有基因Ⅶd亚型(3株)、基因Ⅵ型(2株)和基因Ⅲ型(1株)。这6株NDV的F蛋白裂解位点的氨基酸序列均为112R-R-Q-K-R-F117,属于NDV强毒株典型序列。
Six NDV strains were isolated from chickens and pigeons from Jiangsu province and Henan province. Three virus clones obtained from plaque purification were selected to amplify the F gene by RT-PCR. The full F gene was sequenced and compared with the published sequences of virulent and avirulent strains of NDV in GenBank database. Based on the nucleotide sequence and deduced amino acid sequences, these NDV isolates could be divided into three groups. The group 1 containing 3 NDV strains belonged to genotype Ⅶ, the group including 2 NDV strains belonged to genotypeⅥ and 1 NDV strains in group 3 belonged to genotypeⅢ. The cleavage sites of the F protein of all 6 isolates contain the sequence of ^112R-R-Q-K-R-F^117, a typical motif in virulent strains.
