摘要
目的探讨链脲佐菌素(STZ)对大鼠胰岛β细胞产生一氧化氮(NO)和胰岛素的影响,以及黄芪对胰岛β细胞的保护作用。方法以大鼠胰岛β细胞-RINm5F为研究对象,不同浓度黄芪注射液预处理细胞,STZ诱导细胞破坏,检测培养上清中NO水平;葡萄糖刺激胰岛素释放试验进行细胞功能检测,流式细胞检测细胞凋亡,四甲基偶氮唑盐(MTT)法检测细胞增殖活力。结果10%和20%黄芪注射液预处理组均能显著减少细胞培养液中NO水平,保护β细胞胰岛素分泌、抑制细胞凋亡和促进细胞的增殖。结论黄芪注射液对STZ所致的胰岛β细胞破坏有保护作用,对1型糖尿病具有防治作用。STZ诱导胰岛β细胞凋亡可能与NO介导有关。
Objective To investigate the influence of streptozotocin(STZ) on the production of nitric oxide(NO) and insulin of islet β cells in rats,and the protective effect of Huangqi Injection on islet β cells.Methods The islet β cells-RINm5F in rats were pretreated with Huangqi Injection in different concentrations.STZ was used to induce the lesion of the cells and then the level of NO in islet culture supernatants was detected.The glucose stimulating insulin release test was applied to determine the cell functions,flow cytometry was used to detect the cell apoptosis and MTT method was used to test the cell proliferation.Results Huangqi Injection (10% or 20%) decreased significantly the level of NO in islet culture supernatants,protected the insulin release,inhibited the cell apoptosis and promoted the cell proliferation.Conclusion Huangqi Injection can protect islet β cells from injure by STZ,and prevent and treat type 1 diabetes.The apoptosis of islet β cells induced by STZ may be related to NO mediation.
