荧光光度法测定地表水叶绿素a的探讨 被引量:4

摘要 利用叶绿素a分子具有天然的荧光的性质,采用Cary Eclipse荧光光度计以430nm为激发波长,663nm为发射波长,建立了对地表水叶绿素a含量测定的荧光光度分析方法。叶绿素a浓度在0~150.00μg/L范围内呈良好的线性关系,相关系数r=0.9999,检出限为0.066μg/L,回收率在98%~104%之间。该荧光光度法的灵敏度、准确度和精确度都较好,与传统分析采用的分光光度法进行了比对,结果显示二者并无显著性差异,而且该荧光光度法的操作相比更为简便而灵敏,可以满足并适用于常规地表水叶绿素a的监测。 According to the natural fluorescence inside of chlorophyll - a molecule, a fluorophotometric determination about chlorophyll - a in surface water is created by Cary Eclipse Fluorophotometer with excitation at 430nm and emission at 663nm. Chlorophyll - a concentration has good linear relation in the range of 0 - 150. 00μg/L with correlative coefficient as 0. 9999 and the detection limit of 0. 066μg/L. The recovery is between 98% and 104%. Campared with the tradional method-spectrophotometry, fluorophotometry has no significant difference. With better accuracy sensitivity and precision, it is more eligible to quickly perform the monitoring of chlorophyll -a in surface water.
