桉树种质资源离体保存技术

摘要 利用限制生长法研究桉树(Eucalyptus spp.)种质资源离体保存技术并对保存材料的再生植株进行遗传稳定性检测,结果表明以MS+6-BA0.1 mg/L+NAA0.05 mg/L+蔗糖30 g/L为培养基,在低温(4±2)℃和弱光800 1x培养条件适宜桉树优良种质离体试管保存,保存期6个月,保存成活率达100%。保存6个月的试管苗在增殖和生根培养基上均能正常恢复生长。造林对比试验表明,其再生后代的田间生物学性状与对照植株无差异。运用ISSR标记对再生植株的遗传稳定性进行分析,未发现有遗传变异现象。 Restricting growth methods were used to study the technique of in vitro conservation of Eucalyptus germplasm resources. Genetic stability of regenerated plantlets was also studied. The results indicated that MS + 6 - BA0.1 mg/L + NAA0.05 mg/L + sucrose 30 g/L medium could prolong plantlets' conservation time to six months at (4±2)℃ , 800 1x culture conditions. The survival rate was 100%. After cultured in recovering medium, all plantlets which had been conserved for six months recovered normal growth ability. The afforestation comparative experiment showed that there was no obvious difference in biological characteristics between the regenerated plantlets and control plants, ISSR molecular marker was used to analyze the genetic stability of regenerated plantlets. No genetic variation was detected.
