凋亡抑制基因Livin在宫颈鳞癌组织中的表达及其与Bcl-2的关系 被引量:12

摘要 目的测定凋亡抑制基因Livin在宫颈鳞癌组织中的表达,分析其与凋亡相关基因Bcl-2的关系。方法用RT-PCR方法检测38例宫颈癌组织(均为鳞癌),15例宫颈上皮内瘤变Ⅱ-Ⅲ级(CINⅡ-Ⅲ)组织及16例正常宫颈组织中Livin和Bcl-2 mRNA的表达情况。结果RT-PCR检测结果显示:Livin在宫颈癌组织中的表达与正常宫颈对照组间比较差异有统计学意义(P〈0.05);Livin在宫颈癌组织与CINⅡ-Ⅲ级组织中的表达比较差异无统计学意义(P〉0.05);Livin在CINⅡ-Ⅲ级组织的表达与正常宫颈对照组间比较差异有统计学意义(P〈0.05);Livin的两种异构体α及β均同时表达。Livin的阳性表达与宫颈癌的临床分期比较差异有统计学意义(P〈0.05),与宫颈癌的组织学分级及有无淋巴结转移比较差异无统计学意义(P〉0.05)。另外,在宫颈癌组织中Livin的表达与Bcl-2的表达呈正相关性(r=0.365,P〈0.05)。结论Livin的过表达可能在宫颈癌的发生发展中起一定作用;Livin与Bcl-2的过表达可能在宫颈癌癌变中起协同作用。 Objective To study the expressions of Livin and Bcl-2 in cervical squamous cancer and their relationship. Methods The reverse transcription-polymerase chain reaction(RT-PCR) for Livin and Bcl-2 messenger ribonucleic acid (mRNA) were performed on 38 cases with cervical cancer(squamous cancer), 15 cases with cervical intraepithelial neoplasia Ⅱ -Ⅲ (CIN Ⅱ -Ⅲ ) and 16 cases with normal controls. Results RT-PCR results: there was significant difference in the expressions of Livin between cervical cancer and normal controls ( P 〈 0.05 ). The difference between cervical cancer and CIN Ⅱ -Ⅲ was not significant ( P 〉 0.05 ). There was significant difference in the expressions of Livin between CIN Ⅱ -Ⅲ and normal controls(P 〈 0.05). The expressions of both variants were simultaneous. The expressions of Livin had positive relationship with the clinical stage of cervical cancer ( P 〈 0.05 ), had no relationship with histological grade and lymphatic metastasis (P 〉 0.05). The expressions of Livin were positively related to the expressions of Bcl-2 (r = 0. 365 ,P 〈 0.05 ). Conclusion The overexpressions of Livin may play an important role in process of the pathogenesis and development of cervical cancer. The overexpressions of Livin and Bcl-2 may play synergetic roles in process of carcinogenesis of cervical cancer.
