摘要
目的:通过观察在不同浓度枸橼酸他莫昔芬处理下,大鼠骨髓基质干细胞(rBMSCs)诱导成脂后生长状态的变化,初步探讨用于促进骨髓基质干细胞诱导成脂的最适枸橼酸他莫西芬浓度。方法:培养大鼠骨髓基质干细胞并传代,将第三代的骨髓基质干细胞随机分为三组,分别在加入10-6mol/L,10-7mol/L,10-8mol/L的枸橼酸他莫西芬24h后,进行成脂肪的诱导。分别对三组样本进行四甲基偶氮唑盐检测(MTT检测),测量其光密度值(opticaldensity,OD值)。取第8天的样本进行油红-O染色,计算红色脂滴的数量,并通过多个样本均数的方差分析比较三组之间的差别。结果:①各组OD值之间的差异没有统计学意义(P>0.05);②10-6mol/L组成脂数量明显多于10-7mol/L组和10-8mol/L组,P值均小于0.01;而10-7mol/L和10-8mol/L两组之间在成脂数量上的差异没有显著统计学意义(P>0.05)。结论:10-6mol/L为枸橼酸他莫西芬促进大鼠骨髓基质干细胞诱导成脂的适宜浓度。
Objective To observe the change of OD of rBMSC during differentiating into the adipocytes in the condition of Tamoxifen Citrate with different concentrations,to elucidate the proper concentrations of Tamoxifen Citrate in adipocyte induction. Methods Sprague Dawled rats marrow mesenchymal stem cells were separated and cultivated in vitro. rBMSCs were induced to adipocytes.The induced culture medium was interfered with different Tamoxifen Citrate concentration.MTT, Results There is not significant between different concentrations of Tamoxifen Citrate and adipocytes induction.However, there is a significant between the number of lipids and different Tamoxifen Citrate concentrations.The number of lipids of 10^-6mol/L group is more than other two groups. Conclutions 10^-6mol/L is the proper concentration using Tamoxifen Citrate to effect the adipocyte induction.
