摘要
目的:观察卡维地洛对大鼠心室肌细胞ICa,L电流的抑制作用,探讨卡维地洛抗心律失常的离子通道机制。方法:采用胶原酶酶解法分离大鼠心室肌细胞;采用膜片钳全细胞技术记录大鼠心室肌细胞ICa,L电流,观察细胞外液中加入卡维地洛后,ICa,L电流的变化。结果:对照组及卡维地洛组ICa,L密度分别为(pA/pF):11.25±1.57,3.89±0.59(P<0.05);卡维地洛组ICa,L的I-V曲线向上方移位。结论:卡维地洛能够对抗心律失常的机理之一与它能够阻断ICa,L有关。
Objective :To observe the inhibitory action of carvedilol on L- type calcium channel current (ICa,L) of ventricular myocytes in rats, and to explore the ionic channel mechanism of antiarrhythmic effect of carvedilol.Methods: The ICa,L of ventricle cardiomyocytes isolated by collagenase method was recorded by using patch clamp technique. The influence of carvedilol on ICa,L was analyzed. Results: The ICa,L current densities (pA/pF) were 11.25 ±1.57 in control group and 3.89 ± 0.59 in carvedilol group respectively ( P 〈 0.05 ), and the current - voltage relationship (I- V) curve was shifted upward in carvedilol group. Conclusion:The antiarrhythmic effects of carvedilol may be related with the inhibition of ICa,L in ventricular myocytes.
