摘要
为揭示腰腿痛胶囊治疗腰椎间盘突出症的作用机理,复制了大鼠化学性神经根炎模型,模拟腰椎间盘突出症,分别在造模后3d、7d、14d、21d以全血粘度、血浆粘度为指标,进行血液流变学观察;同时检测神经根周围组织中炎症介质(前列腺素E2,5-羟色胺)水平,并对神经根进行组织学观察。结果是腰腿痛胶囊可明显改善模型大鼠全学粘度、血浆粘度(p<0.05或p<0.01);可显著降低神经根周围组织中的前列腺素E2(p<0.01)和5-羟色胺水平(p<0.01),减轻化学性炎症对神经根的损害。说明腰腿痛胶囊具有活血化瘀作用,可改善化学性神经根炎模型大鼠血液流变学,并具有抗炎镇痛作用,这些作用可能是其临床治疗腰椎间盘突出症的药理机制。
To disclose the mechnism of Yaotuitong capsule treating the lumbar intervertebral disc protrusion,96 wistar rats have been divided into four groups:normal,blank,aspirin and "Yaotuitong capsule"group.Chemical radicular neuritis was duplicated by operation to simulate lumbar intervertebral disc protrusion.Then,viscosity meter was used to detect whole blood viscosity and plasma viscosity at 3,7,14 and 21 days after operation,respectively.Simultaneously,the level of mediators of inflammation was also detected.Morever,histology of nerve root was observed at 7 and 14 day after operation.It is resulted that Yaotuitong capsule could inhibit the abnormal rise of whole blood viscosity,plasma viscosity(p〈0.05 or p〈0.01),significantly depress the level of PGE2(p〈0.01)and 5-HT(p〈0.01)of the local tissue around nerve roots and lessen the injury on the nerve roots.It is conclused that Yaotuitong capsule can activating blood circulation to dissipate blood stasis,has the anti-inflammatory and analgesic effect,which may be one of its pharmacological mechanisms for lumbar intervertebral disc protrusion.
