摘要
目的:探讨草木犀的正丁醇提取物在炎症中的作用。方法:通过LPS干预RAW264.7细胞系建立炎症细胞模型,免疫细胞化学法检验NF-κB的表达及分布变化,实时荧光定量RT-PCR法检测血红素氧合酶1(HO-1)的基因表达,Western blot法测定HO-1的蛋白表达量。结果:免疫细胞化学法结果显示药物干预时胞质被染为棕色,NF-κB多分布在胞质未被激活;仅有LPS干预时,胞核被染为棕色,NF-κB集中分布在细胞核表达增强。药物提取物干预后HO-1的mRNA表达水平明显升高,且呈剂量依赖型关系;Western blot结果显示药物干预后HO-1的蛋白表达水平也明显升高。结论:草木犀的正丁醇提取物通过抑制NF-κB的激活,同时促进HO-1的释放而发挥一定的抗炎作用。
Objective:To study the anti-inflammatory effect of n-butanol extract from melilotus suaveolens ledeb. Methods : Inflammation cell model was constructed by LPS acting on the RAW264. 7 cell line ; the expression and distribution of NF - κB were detected through immunocytochemical method; the expression of mRNA and protein of Heme oxygenase 1 ( HO - 1 ) were detected through real-time PCR and Western blot. Results : The results of immunocytoehemical method showed that cytoplasm stained to brown presented NF - κB inactivation after intervention of the n-butanol extract, cell nucleus stained to brown presented NF - κB activation after intervention of LPS only. The expression of HO - 1 mRNA can be significantly enhanced by the extract in a dose-dependent manner, and the expression of HO - 1 protein can be significandy enhanced too. Concluzion :The n-butanol extract from melilotus suaveolens Ledeb can resist inflammation by inhibi- ting the activation of proinflammatory factor NF - κB and by enhancing the expression of antiinflannnatory factor HO - 1.
