生脉胶囊对压力超负荷心衰大鼠心肌胶原纤维的影响 被引量:1

Effect of Shengmai Capsules on Collagen Fibers of Cardiac Muscle in Rats with Chronic Congestive Heart Failure

摘要 目的:探讨生脉胶囊对慢性心力衰竭大鼠心肌胶原纤维的影响。方法:将大鼠随机分为假手术组、心衰模型组、生脉胶囊组、卡托普利组、生脉胶囊加卡托普利组,采用腹主动脉缩窄法造成大鼠慢性心力衰竭模型,治疗结束时用Masson染色法测定在心肌室胶原容积分数(CVF),免疫组化法结合图像分析仪进行左心室肌Ⅰ型胶原、Ⅲ型胶原含量的测定。结果:①模型组CVF明显高于假手术组(P<0.01),生脉胶囊组、卡托普利组、生脉胶囊加卡托普利组CVF均低于模型组(P<0.01)。②模型组心肌Ⅰ、Ⅲ型胶原的含量均高于假手术组(P<0.05),生脉胶囊组和卡托普利组Ⅰ、Ⅲ型胶原的含量均低于模型组(P<0.01)。结论:生脉胶囊可降低慢性心力衰竭大鼠心肌胶原容积分数和Ⅰ、Ⅲ型胶原的含量,具有减轻心室重构的作用。 Objective : Research the effect of Shengmai capsules on the collagen fibers of cardiac muscle in chronic congestive heart failure(CHF) rats. Methods:75 SD female rats were divided randomly into 5 groups, 15 rats each group: shamoperation group( A), CHF model group (B), CHF model treated by Shengmai capsules group (Shengmai capsule group ,C) ,CHF model treated by Captopril group( Captopril group,D) ,CHF model treated by Shengmai capsule and Captopril group (shengmai capsule & captopril group, E). Suprarenal abdominal artery constriction was operated to prepare CHF rat models. After 7 week treating respectively, the collagen volume fraction (CVF) of cardiac muscle in rats was measured by Masson dyeing and the contents of Col Ⅰ and Col Ⅲ in left ventricular myocardium were analyzed by immunohistochemistry and image analysator. Results:①CVF in group B was significantly higher than in group A (P 〈 0. 01 ), and lower in group C, D,E than in group B (P 〈 0. 01 ). ②The contents of Col Ⅰ and Col Ⅲ in myocardium were both higher in group B than in group A (P〈0. 05) while both lower in group C and D than in group B. Conclusion:Shengmai capsules can reduce the contents of Col Ⅰ, Col Ⅲ and decrease CVF in myocardium, and play a role in relieving the ventricular remodeling of CHF.
