摘要
目的:初步研究山茱萸总苷(TGCO)对实验性变态反应性脑脊髓炎(EAE)大鼠外周血清IFN-γ、sIL-2R的影响。方法:将Wistar大鼠分为3组:对照组、EAE组和TGCO组,采用动物行为学、常规HE和LuxolFastBlue(LFB)染色观察动物的发病情况与中枢神经系统(CNS)的病理变化,ELISA检测血清IFN-γ、sIL-2R的变化。结果:EAE组7/8大鼠出现典型的EAE行为学改变、CNS炎性细胞浸润和髓鞘脱失,血清IFN-γ、sIL-2R值分别是(118.36±12.79,41.17±4.52);而TGCO组有3/8大鼠出现EAE行为学改变,评分最高为2分,潜伏期延长,CNS内炎性细胞浸润和髓鞘明显减轻,血清IFN-γ、sIL-2R值分别是(79.60±17.56,34.86±4.21),与EAE组比较有统计学差异(P<0.05)。结论:TGCO对EAE大鼠有抑制作用,可能与抑制外周血清IFN-γ、sIL-2R有关。
Objective :To primarily analyze immune mechanisms of total glycosides from comus officinalis against rats with experimental allergic encephalomyelitis. Methods : Wistar rats were randomly divided into three groups : Control group, EAE group, TGCC group. The diet, movement, body weight of Wistar rats were daily examined, CNS tissues were stained with Hematoxylin-eosin and Luxol Fast Blue ,and the concentration of IFN -γ, slL- 2R was detected by EL ISA. Results: Seven eithts rats in EAE group demonstrated neurological signs of typical EAE, with parenchymal infiltration and perivascular cuffing with mononuclear cells, and ,demyelination in the white matter, the level of serum IFN -γ, sIL- 2R were 118. 36 ± 12. 79,41.17± 4. 52 individually. However, three eithts rat in TGCO group demonstrated neurological signs of typical EAE,the high score was 2,the latency was prolonged,the infiltration of mononuclear and myelin damages in CNS were reduced, the level of serum IFN - γ, sIL - 2R (79. 60 ± 17. 56,34. 86 ± 4.21 ) were significant lower than that of EAE Group(P 〈0. 05). Conclusion:TGCO could have an effect on prevention on EAE in Wistar rat,may be correlated with its ability to depress the synthesis of serum IFN -γ,sIL- 2R.
