摘要 以带有Tn5的大肠杆菌(Tn5GmlQβλ:F’lac+pro+/△lacprovalr)作为供体。兼性嗜碱细菌NTT89(KmsAmpr)为受体,采用滤膜杂交的方法,然后在含有卡那霉素和氨苄青霉素的平板上得到双抗菌落,可得出Kmr的转移频率远远大于供受体菌的自发突变率,然后把得到的双抗菌落进行筛选,获得5株非嗜碱性突变株。 ? E Coli 142 with Tn5 as donor, facultative alkaliphillic bacillus NTT89 as receptor and with filter hybridization, the colonies which are resistant to Km and Amp on the plates were obtained in the experiment. From these colonies, the unalkaliphilic mutation Bacillus which do not produce on alkaline plates on resistant plates were selected.
