抗人型结核杆菌单克隆抗体的制备和鉴定

摘要 为建立特异、敏感、快速而简单的结核病免疫诊断技术,制备了8株抗结核分枝杆菌的IgG单克隆抗体,经ELISA法与结核分枝杆菌及8株非结核分枝杆菌的PPD鉴定,筛选了其特异性及敏感性。与结核分枝杆菌聚合OT测定了其培养上清抗体滴度在10-2以上。腹水抗体滴度在2×10-4以上,用免疫转印技术确定了与其特异性结合的抗原相对分子质量分别为42.7,27.8KU。8株杂交瘤细胞的染色体计数为98~130条不等。 We produced 8 IgG class specific monoclonal antibodies(McAb)to Mycobacterium tuberculosis for immunodiagnostic purposes. Specificity and sensitivity of 8 McAbs to various PPD of Mycobacteria were developed in ELISA systems. The chromosome numbers of hybridomas were 98 ̄130. Four of the McAbs (TB 20 ,TB 38 ,TB 41 and TB 109 ) were analysed by Western Blot. They showed a specificity restricted to the tuberculosis complex and could be divided into two groups according to their reactivity pattern with two different molecular size: TB 41 , TB 109 reacted with a 27.8 KU molecule and TB 20 , TB 38 reacted with a 42.7 KU molecule.
