载脂蛋白(a)蛋白多态性与皮质脑梗塞相关性研究

Correlative study of apolipoprotein(a) polymorphism with cortical cerebral infarction

目的在蛋白亚基水平上阐明载脂蛋白（ａ）［ａｐｏ（ａ）］多态性与脑梗塞的关系。方法８５名健康对照者和４２例皮质脑梗塞患者以ＥＬＩＳＡ法检测血Ｌｐ（ａ）水平、ＳＤＳ－ＰＡＧＥ／免疫印迹结合银染法检测ａｐｏ（ａ）蛋白大小多态性。结果得出脑梗塞患者血Ｌｐ（ａ）水平显著高于对照组；病人组低分子量ａｐｏ（ａ）表型频率明显高于对照组；但即使是相同ａｐｏ（ａ）表型个体间，病人的血Ｌｐ（ａ）水平仍显著高于对照者。结论研究在进一步证实高水平血Ｌｐ（ａ）是ＡＳ性脑梗塞危险因素的基础上，发现脑梗塞与决定Ｌｐ（ａ）高水平的低分子量ａｐｏ（ａ）表型密切关联，且提示除ａｐｏ（ａ）大小多态性外，尚有其它因素影响血Ｌｐ（ａ）水平。

Objective To study the correlation of apolipoprotein(a)polymorphism with cerebral infarction at the level of protein subtype.Methods The serum Lp(a) level,by means of ELISA,and size polymorphism of apo(a),by SDS PAGE/Western blotting combined with silver staining,were detected in 85 healthy controls and 42 patients with cortical cerebral infarction.Results The serum Lp(a)level of the patients with cerebral infarction was remarkably higher than that of the controls,even between the subjects with the same apo(a) phenotype.The frequency of low molecular weight phenotypes of apo(a) in the patients was also more common than that in the controls.Conclusions At the basis of proving further that the high serum Lp(a) level is a risk factor for atherosclerotic cerebral infarction,this study found the close association of cerebral infarction with low molecular weight phenotypes of apo(a).And it is suggested that there be other factor(s),besides the size polymorphism of apo(a),can affect the serum level of Lp(a).
