摘要
用酵母系统筛选微生物产生的免疫抑制活性化合物,从300多株放线菌中筛选出阳性菌株SIPI-97-84,对其代谢产物进行初步研究,用丙酮浸泡、溶媒萃取、硅胶柱层析和重结晶等方法,从其菌丝体中分离出活性成分SIPI-84-15。通过元素分析和质谱,确定其分子式为C28H31NO6(分子量:477.2145);综合紫外光谱、红外光谱、1H-NMR、13C-NMR等图谱数据,确定SIPI-84-15的结构为含有硝基的多烯化合物,该结构与文献报道的spectinabilin一致,化学命名为:2-甲氧基-3,5-二甲基-6-[四氢-4-[2,4,6-三甲基-7-(4-硝基苯基)-2,4,6-庚三烯亚基]-2-呋喃基],4H-吡喃-4-酮。
Using a method for screening immunosuppressive compounds from microorganisms in a yeast system,a positive strain named SIPI9784 whose metabolites exhibited inhibitory activities on yeasts was obtained from more than 300 Actinomycetes collected in our laboratory.Physiologically active compound SIPI8415 was isolated from the mycelia of Streptomyces sp. 9784 by acetone soaking, solvent extraction, silical gel chromatography and recrystallization etc.. By elemental analysis and MS (FAB), its formula was determined to be C28H31NO6(MW: 477.2145); based on the UV, IR, 1 HNMR, 13 CNMR, HMQC, HMBC etc. spectral data and physicolchemical properties of SIPI8415, the structure of SIPI8415 was elucidated and was identical with that of spectinabilin, its chemical name is 2methoxy3,5dimethyl6[tetrahydro4[2,4,6trimethyl7(4nitrophenyl)2,4,6heptatrienylidene]2furanyl], 4Hpyran4one.
