复方益肾降压口服液对RPH大鼠肾实质保护作用及降压机理研究

摘要 目的探讨中药经验方复方益肾降压口服液对肾实质性高血压(renal parenchymal hypertension,RPH)大鼠肾实质的保护作用和治疗RPH的作用机理。方法通过肾次全切除法制备RPH动物模型,以卡托普利为对照评价该方不同剂量组对模型剩余肾组织形态学和超微结构的影响,并通过免疫组化和RT-PCR法检测模型肾组织内结构型一氧化氮合酶(cNOS)和内皮素-1(ET-1)两种内皮因子的分布、表达。结果复方益肾降压口服液各剂量组均可减轻肾脏病理改变,抑制肾脏局部ETmRNAs表达,并上调cNOS表达。结论复方益肾降压口服液可能是通过抑制肾脏局部ET-1mRNA和上调cNOSmRNA表达、分泌的双相调节作用来实现其降压、保护肾功能的作用。 Aim To explore the protective effects Fufangyishenjiangyakoufuye(FFYSJYKFY) on renal function, a prescription of Chinese herb used for renal parenchymal hypertension ( RPH), and the mechanism of depressurization in RPH rats. Methods 2/3 of the kidney of rats was resected to establish an RPH animal model to investigate the residual renal pathological and ultrastructure effects by light and electron microscope, and the changes of cNOS, ET-1 protein and mRNA expressions by immunohistochemistry and RT-PCR respectively. Results As shown in pathology and compared with control group, FFYSJYKFY groups could reduce the renal pathological changes, ET-1 expression in renal tissue of RPH rat was weakened( P 〈0.01 ) while cNOS expression was enhanced( P 〈0.05) after administration. Conclusion FFYSJYKFY can obviously reduce BP and protect renal function. The mechanism may lie in inhibiting protein and mRNA expressions of ET-1 and simultaneously upregulating those of cNOS.
