摘要
目的对胡椒碱母核结构修饰,并研究胡椒碱及其衍生物的抗疲劳和心肌保护作用。方法胡椒碱及其衍生物(胡椒酸乙酯,胡椒酸甲酯,四氢胡椒碱)的水溶液灌胃给药,通过小鼠力竭游泳实验,测定血浆肌酸激酶,血浆乳酸脱氢酶,血浆尿素氮,血乳酸,血糖等指标;采用异丙肾上腺素损伤原代培养乳鼠心肌细胞,通过MTT法检测心肌细胞存活率,探讨胡椒碱及其衍生物对心肌细胞的保护作用。结果胡椒碱及其衍生物的水溶液能明显的改善小鼠外周血血象,降低小鼠血清中尿素氮、肌酸激酶,血乳酸;其中四氢胡椒碱和胡椒酸甲酯对异丙肾上腺素损伤的心肌细胞有明显的保护作用。结论胡椒碱及其衍生物可以通过其抗疲劳作用和心肌保护作用,提高机体运动机能。
Aim To study the anti-fatigue and protective effects on cardiomyoeyte of piperine and its derivatives by modifying the structure of piperine. Methods Using the exercise exhaustive mice, the treatment group was administered with piperine and its derivatives at different dosages for 7 days. Plasma creatine kinase ( CK), plasma lactate dehydrogenase (LDH), plasma urea nitrogen (BUN), blood lactate (LAC), sugar and other indicators were determined. Then ISO-induced injury of cultured neonatal rat myocardial cells were used to test the protective effect of piperine and its derivatives. Cell viability was analyzed using MTT assay. Results Piperine and its derivatives could reduce the BUN, CK, and LAC concentration in the blood compared with control group, while methyl piperate and tetrahydropiperine could protect against isoproterenol induced neonatal rat cardiornyocytes injury. Conclusion Perprine and its derivatives have anti-fatigue and protective effects on cardimyoyte and can improve the physical function.
