摘要
在有机相双溶剂体系中,用南极假丝酵母脂肪酶B(Novozym 435)催化合成阿魏酸油醇酯,研究了溶剂(logP)、底物分子比、分子筛加量、酶的用量以及反应温度等因素对反应的影响。结果表明溶剂亲脂性越强,底物转化率越高,最适的溶剂为异辛烷/丁酮,最适反应条件为:酸醇比为1:8(n/n),分子筛和酶加量分别为100mg和30mg/mL,反应温度为65℃,反应8d底物转化率最高可达99%。产物阿魏酸油醇酯具有一定抗DPPH自由基的能力,20μmol/L阿魏酸酯的DPPH自由基清除率为34%。
The esterification of ferulic acid (FA) and oleyl alcohol (OA) in organic solvent media by immobilized lipase Novozym 435 catalysis was investigated. The reaction was optimized in terms of selected parameters, including the organic solvent log P, weight of molecular sieve, temperature and substrates ratio (CA/OA).The results showed that the more hydrophobic solvent mixtures,the higher yield of bioconversion.The most appropriate solvent medium was the mixture of iso-octane/2-butanone.The highest esterification yield of 99% was obtained at 65%, a ratio of ferulic acid to oleyl alcohol 1:8,addition of lipase 30mg/mL and lOOmg molecular sieve after 8d.The ester product demonstrated an antiradical activity.
