摘要
本实验对饲料中氨苄青霉素的高效液相色谱(HPLC)检测方法进行了研究。采用磷酸盐缓冲液提取,高效液相色谱-二极管矩阵检测器(HPLC-DAD)检测,在检测波长为220nm的条件下,对饲料进行添加5.0~500mg/kg氨苄青霉素的回收实验。结果表明:氨苄青霉素的平均回收率为76.1%~94.8%(n=6),变异系数为1.7%~7.4%(n=6),检出限为1.0mg/kg,定量限为5.0mg/kg。
The high performance liquid chromatography (HPLC)method was developed for determination of ampicillin in feeds.Samples were extracted with phosphate buffer solution, and detected by high performance liquid chromatographydiode array detection (HPLC-DAD)at 220 nm wavelength.At the level of 5.0 - 500 mg/kg, the mean recoveries of ampicillin ranged from 76.1% to 94.2 %(n=6) ,the coefficient of variance (CV)ranged from 1.7 % to 7.4 %(n=6).The limit of detection(LOD)was 1.0 mg/kg,the limit of quantitation(LOQ)was 5.0 mg/kg.
