迷走神经干切断术后大鼠胃壁细胞DNA含量研究

Research of stomach parietal cell DNA content after vagotomy

目的探讨迷走神经切断术后胃壁细胞DNA含量的变化及机制。方法将健康雄性SD大鼠48只随机分成正常对照组和手术组。手术组大鼠行迷走神经切断术。术后3天至8周取胃壁组织。石蜡切片。HE染色及Feulgen染色。计算机图像分析测定胃壁细胞DNA含量及立体定量。部分标本行透射电镜观察,血清胃泌素测定。结果大鼠胃壁细胞及细胞核的体积减小,DNA含量手术后较对照组增高,壁细胞呈分泌抑制状态。血清胃泌素水平显著增高。结论迷走神经切断术后壁细胞DNA含量增高,可能受高胃泌素血症等神经内分泌因素的调节。

Objective In order to erplore the regulatory mechanism of stomach parietal cell DNA content after vagotomy. Methods 48 SD healthy male rats were randomly divided in to control group (n=8) and treated group. Rats in the treated group undergone truneal vagotomy under intraperitonea aneshthesia. 3days 1, 2, 4, 8 weeks after operation, stomach tissue was taken, 8 rat each times. 5-7cm thick sections were performed and slides stained by HE and Feulgen methods. Stomach parietal cell DNA content was determimea and with a computerized digital imaging analysis system. Areas and diameter of the cells and nuclei were measured at the same time. Some samples were studying un transmission electron microscope, and other samples were used to measure serum gastin. Results The ratsera'stomach parietal cell DNA content was higher after vagotomy than that of control group. And parietal cells were in inhibited state. The level of serum gastrin in treated group was higher than that of control group. Conclusion High level serum gastrin might take part in regulating stomach parietal tell DNA content after vagotomy.
