不规律投料饲养对小鼠糖耐量实验的影响

Effect of irregular breeding on the oral glucose tolerance test in mice

目的观察不规律投料饲养小鼠条件对利用糖耐量实验研究具有α-葡萄糖苷酶抑制活性药物药效学评价的影响。方法将小鼠随机分为常规饲养组(每只小鼠约5g/d)和半量投料饲养组(于糖耐量测定前3d每只小鼠2.5g/d),每个组内又各分为3个处置组,分别灌胃给予阿卡波糖、吐温80和生理盐水,30min后复灌胃给予2.0g/kg剂量的蔗糖溶液,眼眶丛静脉取血测定0、0.5、1.0和2.0h的血糖值,并计算各时间点围成的曲线下面积判断结果。结果半量投料饲养小鼠在糖耐量实验中各时间点血糖值和曲线下面积有不同程度的降低。结论不规律投料饲养条件可对利用在体糖耐量实验研究具有α-葡萄糖苷酶抑制活性药物的药效学评价时产生干扰。

Objective To investigate the effects of irregular breeding on the oral glucose tolerance test （OGTr） in mice for α - glucosidase inhibitors activity study in vivo. Methods The ICR mice were randomly divided into regular breeding group （5.0g/d per mice） and irregular breeding group （2.5g/d per mice three days before test）. There were three different experimental treatment among each group, which were NS negative control, 4% Tween80, and acarbose （25mg/kg） oral administration respectively. The blood glucose level at 0, 0.5, 1, and 2h time point after oral sucrose load were determined. And the area under the curve （AUC） of the blood glucose value at each time point was calculated to evaluate the effects. Results The irregular breeding can reduce the blood glucose concentration and AUC value in OGTT. As the common solubilizer of the lipephilic medicine in pharmacological study, Tween 80 had no effect on OGTT. Conclusion Comparison with the regular breeding, irregular breeding can make disturbance on the evaluation of α - glucosidase inhibitors＇ effect.
