摘要
目的寻找简单实用的高分辨染色体制备及显带方法。方法比较常规制片和秋水仙素与低渗同时处理的方法对提高染色体高分辨的作用,以及常规显带与改良胰蛋白酶磷酸(PBS)显带方法对染色体核型分析结果的影响。结果采用秋水仙素与低渗同时处理进行常规染色体制备,结果显示在5000个分裂相中对照组大于400条带的为482个,占9.64%,实验组为1655个,占33.1%,明显高于对照组(P<0.01);用PBS法进行染色体显带,各条带之间能明显区分,便于染色体核型分析。结论秋水仙素与低渗同时处理可使染色体加长,高分辨染色体出现率明显增高,用PBS法进行染色体显带,操作简单,可明显缩短显带时间。
Objective To study and find a simple and pragmatic method for chromosome preparation and banding. Methods The effect of routine chromosome preparation was compared and treated by colchicines, normal chromosome banding and changed trypsin banding for chromosome analysis. ResultsThe chromosomes were longer than before after treated by colchicines, banding with changed trypsin can reduce the time. Conclusion Treated by colchicines and changed trypsin is a simple and pragmatic method for chromosome preparation and banding.
