摘要
目的:探讨FHL2对Id2蛋白功能的调控效应。方法:将E47、5xE-Box-Luc、pRL-SV40、Id2以及FHL2表达载体分别共转染MCF-7、293T以及SH-SY5Y细胞,双荧光素酶报告基因方法检测不同转染组细胞中的相对荧光素酶活性。结果:三个细胞系中E47均显著上调了报告基因的表达,Id2抑制了E47的转录激活活性,而FHL2通过与Id2相互作用显著消弱了Id2对E47功能的抑制效应。结论:通过相互作用FHL2抑制了Id2蛋白的功能活性,FHL2是一个新的Id2蛋白功能抑制子。
Objective: To study the role of FHL2 in regulation of Id2 functional activity. Methods: Expression vectors including FAT, 5xE Box Luc, pRL SV40, Id2 and FHL2 were cotransfected with MCF-7, 293T and SH-SY5Y cells, respectively. Relative luciferase values for transfected cells were measured by dual luciferase report assay. Results: FAT significantly upregulated the expression of reporter gene in three cell lines. Id2 inhibited the activity of FAT. FHL2 decreased the inhibitory effect of Id2 by interacting with Id2. Conclusion : FHL2 is a novel functional repressor of Id2 by suppressing the functional activity of Id2.
