摘要
目的:探讨藤茶总黄酮(TF)对免疫抑制小鼠细胞免疫功能的影响。方法:以皮下注射地塞米松或环磷酰胺(CTX)复制小鼠免疫抑制模型,药物组给予不同剂量的TF(200,100和50mg·kg-1),并设空白组与模型对照组。分别以5,10μg·mL-1刀豆蛋白(ConA)刺激脾淋巴细胞,以MTT法测定其增殖功能;以MTT法和廓清率测定小鼠的NK细胞杀伤活性和单核细胞吞噬功能。结果:TF能够显著提高免疫抑制小鼠NK细胞杀伤活性和单核细胞廓清率。当ConA浓度为10μg·mL-1时,200mg·kg-1和100mg·kg-1TF能促进免疫抑制小鼠脾淋巴细胞增殖;当ConA浓度为5μg·mL-1时,200mg·kg-1TF能显著促进免疫抑制小鼠脾淋巴细胞增殖。结论:TF可提高免疫抑制小鼠的细胞免疫功能。
Objective: To investigate the immunomodulation of total flavonoids of ampelopsis grossedentata (Hand-Mazz) W.T. Wang (TF) on immunosuppressed mice. Methods: The immunodepressed mice were established by means of subcutaneous injection of dexamethasone or cyclophosphamide (CTX). The mice of administration groups were given TF doses of 200, 100 and 50 mg·kg^-1 respectively in comparison with the control group and the model group. MTT method was used to detect lymphocyte proliferation induced by ConA (5, 10 μg·mL^-1). The killing activity of NK cells and the phagocytosis of monocyte were tested by MTT method and clearance rate. Results: TF can significantly increase the killing activity of NK ceils and clearance index of monocyte in CTX-treated mice. 200 and 100 mg·kg^-1 dose of TF could enhance lymphocyte proliferation with ConA (10 μg·mL^-1); 200 mg·kg^-1 dose of TF could significantly enhance lymphocyte proliferation with ConA (5μg·mL^-1). Conclusion: TF can improve cell-mediated immunity of immunosuppressed mice.
