摘要
以引自日本村田种苗公司的菜用羽衣甘蓝杂交种‘绿叶多汁’为试材,对小孢子胚诱导条件、胚状体转接时间、生芽培养基和生根培养基进行了优化筛选。在NLN-13+0.1mg·L-16-BA+0.1mg·L-1NAA和NLN-13+0.2mg·L-16-BA+0.2mg·L-1NAA两种培养基上获得了胚状体,其发生率分别为1.38胚.蕾-1和0.98胚.蕾-1;转胚时间以接种培养后25d为宜;适宜的胚状体生芽培养基为MS+2.0mg·L-16-BA+0.1mg·L-1NAA+3%蔗糖+0.7%琼脂,丛生芽诱导率为56.38%;生根培养基以1/2MS+0.1mg·L-1NAA+3%蔗糖+0.7%琼脂为宜,生根率100%。
Plant regeneration was firstly reported by means of isolated microspore culture in edible kale. A hybrid cultivar named ' Succulent Green Leaf' which was introduced from Japan was used for the experi- ment. Several combinatory were screened in microspore-derived embryos, trans-embryos time, hormone con- stitution on shooting and rooting in order to obtain the best way on the regenerative system. Embryos were attained in the mediums of NLN-13 +0. 1 mg·L^-1 6-BA +0. 1 mg ·L^-1 NAA (embryo yield per bud was 1.38) and NLN-13 +0. 2 mg·L^-1 6-BA +0. 2 mg·L^-1 NAA (the yield was 0. 98). The suitable time for transfer of the embryos was 25 days after inoculation; The best shooting medium was MS + 2. 0 mg ·L^-1 6-BA +0. 1 mg·L^-1 NAA +3% sugar +0. 7% agarose. With the shoot induction rate was 56.38%, the suitable medium for rooting was 1/2MS +0. 1 mg ·L^-1 NAA +3% sugar +0.7% agarose and the root inducing rate come up to 100%.
