白花蛇舌草提取液诱导多药耐药白血病细胞HL-60/ADR凋亡的研究 被引量:2

摘要 目的:研究白花蛇舌草乙酸乙酯提取液对多药耐药白血病细胞HL-60/ADR的抑制作用及其机制。方法:采用MTT比色试验观察白花蛇舌草乙酸乙酯提取液对多药耐药细胞HL-60/ADR的抑制作用(HL-60/ADR的耐药倍数为204倍)。采用光镜、电镜技术观察细胞形态结构的改变,利用流式细胞术(FCM)检测细胞凋亡率。结果:白花蛇舌草乙酸乙酯提取液能明显抑制HL-60/ADR细胞的生长,药物浓度相当于生药41.67mg/ml时,即具有显著的抑制作用,抑制率呈剂量和时间依赖性,半数抑制浓度(IC50)相当于生药70.41mg/ml。流式细胞学证实白花蛇舌草乙酸乙酯提取液,能诱导多药耐药细胞HL-60/ADR凋亡,凋亡率也呈时间和剂量的依赖性。结论:白花蛇舌草对多药耐药细胞HL-60/ADR的生长具有极强的抑制作用,诱导其凋亡是其机制之一。 Objective : To study the inhibitory action and mechanism of acetic acid ethyl ester extracts from Hedyotic diffusa on muhidrug resistance leukemic cell line HL-60/ADR. Methods : To observe the depressant effect of acetic acid ethyl ester extracts from Hedyotic diffusa on muhidrug resistance leukemic cell line HL-60/ADR (the drag-fast multiple of HL-60/ADR is 204)by MTF. To observe the change of the cells morphology structure by light microscope and electron microscope, to detect the apoptosis rate by flow cytometry (FCM). Results : The extracts from Hedyotic diffusa could effectively inhibit the proliferation of HL-60/ADRcells. When the concentration is amount to 41.67mg/ml in pharmacognostics, it has effectively inhibitory action. The inhibition rate has dependence of dosage and time. The IC50 is amount to 70.41 mg/ml in pharmacognostics. Acetic acid ethyl ester extracts from Hedyotic diffusa could induce apoptosis of muhidrug resistance leukemic cell line HL-60/ADR have been confirmed by flow cytometry(FCM) ,the apoptotic rate has dependence of dosage and time. Conclusion: Hedyotic diffusa could effectively inhibit the proliferation of muhidrug resistance leukemic cell line HL-60/ADR,induce HL-60/ADR to apoptosis is one of its mechanism.
