摘要 目的:探讨KI治疗孢子丝菌病的机理。方法:通过CFU计数观察不同组抗菌能力的差异。用DCFH-DA标记被申克孢子丝菌刺激后的PMNs,激光共聚焦检测PMNs内ROS的水平。结果:单独的PMNs组的CFU计数明显少于单独的Lym组或MNC组(P<0.01),而同时含有PMNs和Lym的实验组的CFU又明显少于其他组,差异有显著性(P<0.01)。在有无Lym参与作用下,其PMNs内的ROS水平都无明显差异(P>0.05)。结论:KI可以通过活化Lym后表达某些细胞因子来促进PMNs杀菌,但其杀菌机制与ROS无关。 Objective: To investigate the anti- fangus mechanism of KI against S. schenckii. Methods: Count the numbers of CFU used to explore the differenee of anti - S, schenckii ability. Detect the pnxtuction of ROS of the S. schenckii - stimulated PMNs which labeled with DCFH - DA measarod by laser confocal microscopy. Results: The numbers of CFU of/he group which only contains PMNs is less than only contains Lyre or MNC ( P 〈 0.01), and the numbers of CFU of those groups which contain PMNs and leukomonocytes ( Lym) are less than other groups, the difference has prominent significance ( P 〈0.01 ). No matter have Lym or not, there is no obvious difference of the level of ROS in PMNs between the two groups ( P 〉 0.05). Conclusion : KI can activate Lyre to ~crete some cylokine and it can promote the anti - S. Schenckii ability of PMNs, but not by regulate the level of ROS in PMNs to implement the effect.
