摘要
目的研究尼莫地平对大鼠脑缺血/再灌注损伤治疗时间窗的影响。方法将60只成年雄性Wistar大鼠随机分成,尼莫地平组和对照组两大组。每个大组再分成5个小组,即MCAO2h,3h,4h,5h和6h组。应用线栓法制作大鼠MCAO模型。于再灌注前20min、再灌注后12h和36h尾静脉给药;48h后计算大鼠存活率、行神经功能缺损评分、脑血流量和脑梗死体积测定。结果①大鼠存活率:尼莫地平组存活率为53.3%(16/30只),对照组为40%(12/30只)。大鼠存活率提高,但没有统计学意义。②神经功能缺损评分:尼莫地平组神经功能缺损明显少于对照组。其中MCAO4h的大鼠神经功能尼莫地平组明显好于对照组(P<0.05)。③脑组织血流量:尼莫地平组的大鼠脑血流量明显大于对照组(P<0.05)。各时段大鼠脑血流量,尼莫地平组也明显大于对照组,其中MCAO2h,3h和4h有统计学意义(P<0.05)。④梗死体积:尼莫地平组的大鼠脑梗死体积明显小于对照组,其中MCAO2h组的大鼠脑梗死体积减小最突出(P<0.05)。结论尼莫地平对大鼠脑缺血/再灌注损伤模型的治疗时间窗的影响在2h~4h范围。
Objective To investigate the effects of the therapy of nimodipine in the time window for reperfusion injury after focal cerebral ischemia in rats. Methods Sixty adult male Wistar rats were randomly divided into two groups, nimodipine, and control group. The nimodipine and control group once more divided into 2h, 3h, 4h, 5h, and 6h groups. A rat model of middle cerebral artery occlusion (MCAO) was induced by the intraluminal suture methods Reperfusion was performed each time stage after cerebral ischemia, respectively. At 20 minutes before reperfusion after cerebral ischemia, 12 h and 36 h after reperfusion, the medications were injected slowly into the tail veins of rats. The survival rate of rats was calculated 48 hours after cerebral ischemia/reperfusion, and the volumes of cerebral infarction and neurological deficit scores were measured. Results①The survival rate of rats in the two groups were 53.3% and 40%, respectively. The survival rate in nimodipine group is higher than that in the control group. But it was not statistical significance.②Neurological deficit scores: Neurological deficit scores in nimodipine group were less than the control group. And the nimodipine 4h group were less than the control group (P〈0.05).③Cerebral blood flow in rats: The nimodipine group has a higher cerebral blood flow (P〈0.05). And the 3h with 4h groups are higher cerebral blood flow (P〈0.05).④The volumes of cerebral infarction in rats: The volumes of cerebral infarction in nimodipine group was less than control group and 2h group has statistical significance (P〈0.05). Conclusion The neuroprotective effects of nimodipine in the time window on focal cerebral ischemia/reperfusion injury in rats were 2h to 4h.
