摘要
目的:探索异体组织工程牙胚移植的免疫反应,明确诱导免疫抑制药物他克莫司对免疫相关因子的影响。方法:采用出生后猪第一磨牙牙胚与煅烧骨复合构建组织工程牙胚植入异体猪肠系膜,移植后应用他克莫司作为免疫抑制剂,特定时间点取材,HE染色观察移植牙胚的发育及免疫反应,免疫组织化学及图像分析了解免疫相关因子CD4/8的表达变化。结果:肠系膜移植后移植物内可见大量浸润的淋巴细胞,应用他克莫司后,CD4+T细胞的分泌明显下降,组织工程牙胚可在大网膜内进一步发育形成牙髓-牙本质复合体。结论:他克莫司可有效地避免因牙胚异体移植导致的免疫反应,本试验为以后组织工程牙胚的临床应用提供了实验基础。
Objective:To investigate the immune response of porcine transplanted with allogeneic tissue-engineered tooth germ and the influence of CD4/8 with inductive immunosuppressive drugs FK506. Methods Porcine tooth germ cells and ceramic bovine bone were used to construct tissue-engineered teeth and the mixture was implanted into mesenteries. Immunosuppressant FK506 was administrated after implantation. At special time points, the implants were retrieved for histological analysis.Results:There were obvious lymphocyte infiltrations in the graft after implantation of mesenteries. The levels of CD4+T cells were significantly decreased and the further growth of allogeneic tissue engineered tooth germ in vivo was obvious after administration of FK506. Conclusions:Immunosuppressant FK506 can affect the immune response caused by the implantation of allogeneic tissue-engineered tooth germ. These studies provided a promising experimental foundation for the clinical application of tissue engineered tooth germ in future.
