参附注射液对失血性休克大鼠内皮细胞C蛋白受体表达的影响 被引量:1

Effects of Shenfu injection on expression of endothelial protein C receptor in hemorrhagic shock: experiment with rats

摘要 目的探讨失血性休克时内皮细胞C蛋白受体(EPCR)的蛋白质和mRNA表达的变化以及参附注射液的干预作用。方法30只Sprague Dawley大鼠随机均分成3组:休克组(经放血建立失血性休克模型,不予复苏),Ringer氏液复苏组(建立失血性休克模型并稳定60min后用3倍失血量的Ringer氏液复苏),参附复苏组(建立失血性休克模型并稳定60min后用3倍失血量的含参附注射液10ml/kg的Ringer氏液复苏)。另外10只SD大鼠仅接受假手术,作为对照组。复苏2h后,处死大鼠取出肝脏。Western印迹法测EPCR蛋白质水平,RT—PCR法测EPCRmRNA水平。结果对照组可测得EPCR蛋白质和mRNA表达。休克组和Ringer氏复苏组的EPCR蛋白质表达显著低于对照组,而EPCRmRNA表达显著高于于对照组(均P〈0.01)。休克组和Ringer氏复苏组的EPCR蛋白质和mRNA表达间无显著性差异(均P〉0.05)。参附复苏组EPCR蛋白质表达显著高于休克组和Ringer氏复苏组,而EPCRmRNA表达显著低于休克组和Ringer氏复苏组(均P〈0.01);参附复苏组的EPCR蛋白质和mRNA表达与对照组相比无显著性差异(均P〉0.05)。结论失血性休克时EPCR蛋白质表达下调,而EPCRmRNA表达上调。参附注射液通过对内皮细胞的保护作用,具有抗休克作用。 Objective To investigate the effects of Shenfu injection containing roots of ginseng and monkshood, traditional Chinese medicinal herbs, on the protein and mRNA expression of endothelial protein C receptor (EPCR) during hemorrhagic shock. Methods Thirty Sprague Dawley rats were made hemorrhage shock models by depletion of blood and then were randomly divided into 3 equal groups: hemorrhagic shock group (HS Group) without resuscitation, Ringer's solution resuscitation group (RS Group), undergoing perfusion of Ringer's solution with the volume 3 times that of the blood loss 60 min after the post-depletion condition became stable, Shenfu injection resuscitation group (SF group) undergoing perfusion of Ringer' s solution mixed with Shenfu injection 10 ml/kg with the volume 3 times that of the blood loss 60 rain after the post-depletion condition became stable. Another 10 rats without depletion of blood qwer used as control group (C Group). Two hours after being resuscitated, the rats were executed with their livers taken out. Western blotting was used to detect the protein expression of EPCR and reverse-transcription polymerase chain reaction was used to measure the EPCR mRNA expression. Results Both protein expression and mRNA expression of EPCR could be found in C Group. The EPCR protein expression levels of HS and RS Groups were both signigficantly lower than that of C Group, and the EPCR mRNA expression levels of HS and RS Groups were both obviously higher than that of C Group (all P〈0.01). There were not significant differences in the protein and MRNA expression levels of EPCR between HS and RS Groups (both P〉 0.05). The EPCR protein expression level of SF Group was significantly higher than those of HS and RS Groups, and the EPCR mRNA expression level of SF Group was significantly lower than those of HS and RS Groups (all P〈0.01). There were not significant differences in the EPCR protein and mRNA expression levels between SF and C Groups (both P〉0.05). Conclusion The EPCR protein expression is down-regulated, and the EPCR mRNA up-regulated significantly in hemorrhagic shock. Shenfu injection plays an anti-shock role, perhaps, by protecting the endothelial cells from injury.
